Overview

[11C] raclopride is a radiolabeled form of the dopamine D2 receptor antagonist raclopride, in which the molecule is tagged with the radioactive isotope carbon-11. It acts as a selective antagonist at dopamine D2 receptors, with much lower affinity for D3 and negligible affinity for other dopamine receptor subtypes. The compound is primarily used as a positron emission tomography (PET) radiotracer to non-invasively image and quantify striatal dopamine D2 receptor availability in vivo. This allows assessment of dopaminergic function in neurological and psychiatric disorders such as Parkinson's disease, Huntington's disease, schizophrenia, major depressive disorder, and substance use disorders. The binding of [11C] raclopride can be displaced by endogenous dopamine or pharmacological agents that increase synaptic dopamine levels; thus it serves as an indirect marker for changes in extracellular dopamine concentrations during various tasks or drug challenges[2][3][4][5][6][8].
